He was a partner in the successful wholesale druggists of W.H. Schieffelin & Fowler, started by his grandfather, Jacob Schieffelin III. Henry was "a man of large benevolence and especially interested in the advancement of negroes". He had been Consul-General in Liberia and was a founding member and President of the New York Colonization Society who financed a mission to explore the interior of Liberia. He supported recruiting African-American students to enroll at Liberia College and encouraged emigration. He lived at 665 Fifth Avenue, a five story house in Manhattan, and kept a country home "Ashton" in Yonkers, New York. He died at Alexandria, Egypt, where he had gone for the benefit of his health.
